Utilización de bolsas ANKOM® en la determinación de la digestibilidad de la materia seca in vitro en conejos.

The aim of this work was to compare the use of ANKOM® bags vs. crucibles to determine in vitro dry matter digestibility (ivDMD, performed in two or three steps corresponding to ileal and faecal digestibility, respectively) of four rabbit diets and six raw materials with a wide range of DM digestibility (from beet pectins to lignocellulose). Crucibles were compared to bags placed collectively (up to 30 in a 3.6 L Daisy jar in a rotating in vitro incubator system Daisy-II). Once digested, samples placed collectively were washed with water (40ºC, 30 min, rotating in Daisy system, and additionally rinsed two times 5 min. in water), etanol and 96% acetone (each one for 5 min.). Bag samples were corrected with blanks. Each analysis was done at three different times being replicated twice each time and using the average of each time and feed for statistical analysis. Values obtained for ivDMD in two steps were similar using crucibles or bags for the whole feeds (P = 0.44, 43.1 vs. 42.8%, respectively), with no interaction feed x method. However, ivDMD in three steps decreased in collective bags compared to crucibles (- 0.6 percentage units. P = 0.009). This narrow difference is probably detected due to the reduced variability observed in this determination. Besides, an interaction feed x method was found for three steps ivDMD (P = 0.002), as digestion in collective bags was lower than that of crucibles for sugar beet pulp, sunflower hulls and straw, with no effect on the other feeds and in none diet. In general, these differences were small, ranging between 2.0 to 2.6 percentage units. Correlation between two and three step ivDMD with crucibles and bags was very high (r ? 0.98. P < 0.001). These results suggest that ANKOM® bags can be used as an alternative to crucibles in ivDMD.
