We will deliver at least one model plant or plant cell culture (Tobacco/Common Bean) in which modules 1 and 2 are functional in at least one spatial (organelle) and temporal (constitutive/inducible) condition. A preliminary assessment of the metabolic bargain of overexpressing Nif components in plants will also be provided by using commonbean as a model. These model systems are used to establish faster high-throughput protocols yielding enough biological material for subsequent biochemical assays. The key result is the validation of the ability of plastids to host active nitrogenase components. The project includes three objectives: (i) generate three multi-gene constructs for modules 1 and 2 avoiding silencing events; (ii) assess the functionality of modules 1 and 2 in chloroplasts from 5 independent transformants; and (iii) Assess the functionality of modules 1 and 2 in amyloplasts from 5 independent transformants. | |
