Molybdenum is an essential micronutrient required in Symbiotic Nitrogen Fixation (SNF). It is a key component of the Fe-Mo-cofactor used by nitrogenase to convert N2 into NH4+. In the model legume Medicago truncatula, MtMoT1.3 is the member of the MoT1 family1 with the highest expression levels in nodules. Yeast complementation tests reveal that MtMOT1.3 transports molybdate into cytosol. MtMOT1.3 is located in the plasma membrane in nodule cells, as inmunolocalization studies show. A loss-of-function mot1.3-1 mutant exhibited reduced growth and nitrogenase activity compared to the wild type under symbiotic conditions, but not when plants are fertilized with inorganic nitrogen. Mot1.3-1 mutant phenotype is restored to wild type levels either irrigating with high concentration of molybdate solution or by complementing it with the MtMOT1.3 wild type allele. These data show up the first molybdate transporter described involved in SNF. This work is a first step towards the understanding of Mo homeostasis related to SNF. | |
