Gibberellins (GA) are plant hormones that control plant growth and development by regulating gene expression differentially accros tissues and their levels can also be altered in response to environmental cues. In Arabidopsis seeds, GAs are required for germination by promoting embryo growth and weakening of the endosperm cell walls (Holdsworth et al., 2008). We have defined the molecular mechanism coordinating growth in the embryo (Rombolá-Caldentey et al., 2014) and we have recently found that endosperm cells also grow during seed germination. By using complementary laboratory and in silico approaches (Castrillo et al., 2011; Bassel et al., 2011), we have identified 6 transcription factors (TF1 to TF6) that differentially regulate the transcription of a selected marker gene for this process (EGM). The binding sequence for two of these TFs (TF1 and TF2) in the EGM promoter has been determined and our results indicate that: i) Both TFs integrate GA-signalling in the seed endosperm and are positive regulators of EGM expression; ii) Their role in this regulation is not completely overlapping since single loss of function (KOs) mutants have delayed germination kinetics and reduced EGM expression; iii) They control the expression of several genes putatively involved in endosperm cell growth. Our ongoing research is expanding on these results and trying to determine to what extent this mechanism has been recruited by other developmental stages and/or plant species to control growth, as well as studying the integration of other inputs (mainly TF3 to TF6 and environmental signals) into this regulation. | |
