Symbiotic nitrogen fixation (SNF) carried out by endosymbiotic rhizobia in legume nodules requires of Molybdenum (Mo) as a component of the Iron-Molybdenum cofactor (FeMoco) of nitrogenase. This bacterial enzyme is responsible of fixing atmospheric nitrogen into ammonia, which is transferred to the host plant in exchange of photosynthates and mineral nutritents, Mo among them. We hypothesize that Mo is delivered by the vasculature to the apoplast of the nodule cortex, where it will be incorporated by the rhizobia-infected nodule cells. MtMot1.2 is a Medicago truncatula belonging to the MOT1 family of molybdate transporters, candidate to being responsible for Mo delivery to the nodules. This transporter is expressed in nodules and roots of nodulated plants. Inmunolocalization assays indicate that it is located in the perycicle and endodermis of the vessels in roots and nodules. Loss-of-function insertional mutant, mot1.2-1, showed a reduced nitrogenase activity which cause a light reduction in the growth. No significant difference in growth was observed between mot1.2-1 and WT when ammonium nitrate is provided in the nutritive solution as nitrogen source. | |
