Characterization of American strains of Colletotrichum tofieldiae

The fungal endophyte Colletotrichum tofieldiae (Ct) promotes plant growth and silique production under phosphate starvation conditions. The mutualistic interaction was originally observed with isolates from asymptomatic wild Arabidopsis thaliana plants growing at a natural site at Central Spain. In this work, we have isolated new strains of C. tofieldiae from surface-disinfected roots, leaves, stems or siliques of asymptomatic A. thaliana plants from a wild population in North Carolina (USA). To characterize the effect of these new strains on plant performance, we conducted different assays using A. thaliana Col-0 plants grown either in vitro (MS medium) or in poor-nutrient soil. In the in vitro assay, two American Ct strains were compared with the reference Spanish strain to study root development and biomass production. Results showed that the two American and the Spanish Ct strains displayed a similar behaviour, increasing the root length and the fresh weight of the rosette leaves compared to mock inoculated plants. In the in soil assay, four American strains and two Spanish Ct strains were compared by measuring the total number of siliques at the end of the cycle. All Ct strains, both the American and the Spanish ones, increased the number of siliques produced by the plant. These results indicate that the beneficial effect of Ct in A. thaliana Col-0 does not vary significantly with the geographical origin of the isolates.
