The immune response to epizootic rabbit enteropathy (ERE) was assessed through the study of pro- and anti-inflammatory cytokine gene expression profiles in healthy and diseased rabbits. Appeared in 1997, ERE is still nowadays one of the main severe intestinal disorders in rabbit farms in Europe. Characteristic symptoms of this disease usually appear two weeks after weaning however there is a lack of studies describing the immune response associated to the disease. In the present study real time RT-PCR analysis was used to determine the expression of IL-2, IL-6, IL-8, IL-10, IFN-¿, TNF-¿ e iNOS in diseased relative to healthy rabbits. To this end eight crossbred (New Zealand White × Californian) rabbits obtained from a farm affected by ERE were used. Animals weaned at 25-d-old and weighing 473 ¿} 8 g had free access to a commercial feed and ad libitum access to water throughout the experimental period. At 39 d of age four healthy animals and four animals displaying ERE symptoms were slaughtered and 3cm ileal mucosal scrapings were sampled and stored in RNA later. After total RNA isolation the expression of selected cytokines was assessed by RT-PCR and the fold change in gene expression of diseased animals relative to healthy evaluated by the 2DDCt analysis using HPRT as housekeeping gene. A significant (P < 0.01) gene expression increase was observed for IL-6, IL- 8, IL-10, iNOS and TNF-¿ in rabbits affected by ERE. The larger fold-differences in expression between the diseased and normal animals were observed for IL-6 (41.9-fold increase), IL-8 (24.1-fold increase), iNOS (20.1-fold increase) and TNF-¿ (11.39-fold increase). Given the high expression of inflammatory cytokines and iNOS in diseased animals it is concluded that rabbits affected by ERE display a cell mediated immune response.
