Seeds from Quercus species show different degrees of recalcitrance; this characteristic limits their conservation time and their availability. Quercus species play an important role in the different habitats of the Iberian Peninsula. Holly oak (Q. Ilex) and cork oak (Q. suber) forests are the most characteristic formations of the dry areas of the Peninsula. In vitro shoot cultures of Q. ilex were initiated from 2-year plants and from seedlings. Best initial proliferation results (85%) were obtained from juvenile material. Experiments thereafter were carried out with this type of material. In the multiplication phase, best proliferation results were obtained on WPM medium (Woody Plant Medium; Lloyd & McCown, Combined Proceedings International Plant Propagation Society 30, 421-427, 1981) + 1 mgL-1 BA (benzyladenine) out of the nine combinations of growth regulators tested (92% proliferation, 4.5 shoots/explants). Rooting proved difficult and only 35% shoots elongated roots (best results, obtained on 1 mgL-1 IBA, indole-3-butyric acid). After 6 months of in vitro conservation of Q. suber shoots (without subculture or transferring to fresh medium), best results were obtained at 4 ºC in darkness on WPM+ 0.1 mgL-1 BA. These methods contribute to germplasm conservation of these species, together with cryopreservation procedures developed. | |
