The diversity and genetic structure of a collection of 190 Spanish durum wheat (Triticum turgidum L) landraces has been assessed by using SSRs, DArTs and gliadin markers. The genotypes analysed were separated into nine populations with a moderate to great genetic divergence among them. The three subspecies taxa, dicoccon, turgidum and durum, present in the collection, largely determined the clustering of the populations, with ssp. turgidum showing an intermediate genetic distance from durum and dicoccon. DArT markers revealed two geographic substructures for dicoccon (east-west) and for turgidum (northeast-southwest). Agro-morphological traits and gliadin composition showed that these geographic patterns resulted from adaptation to contrasting environments. The ssp. durum had a more complex genetic structure, consisting of seven populations with high intra-population variation. Within some of these populations, the analysis of DArT markers discriminated subgroups with agro-morphological and gliadin differences, and distinct agro-ecological zones of origin. Two different phylogenetic groups were detected, as some durum populations appear to be closer to the ssp. turgidum landraces from northern Spain, while others, markedly different, less diverse and more isolated, seem to be more related to durum wheats from North Africa. A large subsample of durum and turgidum landraces has been evaluated for mineral (iron and cinc) content in whole grain. The analysis has evidenced a wide variability, as well as the potential of some genotypes to be incorporated into breeding programs for enhancing durum wheat nutritional quality. | |
