Next-generation sequencing of pooled DNA samples (Pool-Seq) is a cost-effective, attractive approach to study Rhizobium population genomics. We have established a data processing pipeline to align Pool-Seq datasets against a reference genome and to visualize and analyze read coverage and polymorphic sites (SNP) distribution in the population. This pipeline has been used to test the hypothesis ?and show at the genomic level? that different plant hosts (Pisum sativum and Vicia faba) of Rhizobium leguminosarum bv. viciae select specific genotypes from those present in a given soil. Advantages and limitations of the use of Pool-Seq for Rhizobium population genomic studies are also discussed. | |
