The pericycle tissue gives rise to lateral root founder cells (LRFC) through a reprogramming process, and subsequently, distinctive cell fates are specified trough asymmetrical divisions. Morphogenesis of lateral roots initiates with the asymmetric division of LRFC to generate small and large cells. These divisions require external inputs (auxin hormone) and are driven by intrinsic cues (such as polarity and nuclear migration). We hypothesize that self-organizing properties of founder cells are controlled by a regulatory network which incorporates external cues such as auxin. Trough double Fluorescent Activated Cell Sorting we will be able to know the expression levels of genes in pericycle, lateral root founder cells and its daughters. To these end, we have already generate a range of plants carrying cells markers: a) pericyle cells capable of undergoing reprogramming will be isolated using the line carrying the markers J0121_pSKP2B0.5:ER::3xmcherry, b) pericyle cells undergoing reprogramming trough the line carrying DR5:D2eGFP::eGFP_pSKP2B0.5:NLS::3xmcherry, c) founder cells trough the marker line pWOX5:FP_pSKP2B0.5:NLS::3xmcherry, and the LRFC daughter cells will be isololated using d) the line carrying the markers pWOX5:FP_pSCR:ER::3xmcherry for the small daughter cell), and e) the markers pWOX5:FP_pHB53-2K:ER::3xmcherry for the large daughter cell. This approach will define the regulatory program between crucial developmental states (pericycle, lateral root founder cells and its daughters) associated to root organ morphogenesis addressing how two distinct fates are specified from a single cell. We except that our approach provides novel relationships between pluripotency and cell identity. | |
