Winter dormancy, a developmental program evolved in perennial trees from temperate and boreal latitudes to survive under unfavorable conditions during the winter, involves meristematic growth cessation together with morphological and physiological changes required to avoid plant cell freezing (1). Interactions between photoperiodic and temperature signaling pathways with the endogenous developmental programs are essential to define the growth-dormancy annual cycles. A coincidence mechanism, between the internal clock and photoperiod, initiates dormancy entry. When daylength is shortened below a critical value, so called the critical daylength (CDL), perennial tree species stop growing and set buds (2). Therefore, understanding the CDL mode of action has great biotechnological interest because directly impacts in tree productivity and helps to understand how tree will respond to future global climate change. The circadian clock, as well as the module CONTANTS / FLOWERING LOCUS T, emerges as a part of CDL regulation. Nevertheless, additional elements are needed to understand CDL mechanism (3). In this work we present a circadian clock-controlled RAV1 transcription factor ectopic overexpressed in poplar that delays dormancy entry under SD conditions. In addition, RAV1, which acts as a repressor, affects clock gene expression when it is transiently overexpressed in pLHY2:LUC poplar plantlets. Moreover, we set up a RAV1 inducible system in poplar to unravel its primary targets genes. As a result, we found that LHY2 is a RAV1 target gene and that RAV1 could be participating in CDL regulation during dormancy entry. | |
