Astragalus nitidiflorus is an endangered and endemic legume of SE Spain. After was considered extinct, it was re-discovered in 2004 and later included in a governmental conservation program. The main objective of the project is the reintroduction and maintenance of the A. nitidiflorus. According to this purpose, the isolation and characterization of its endosymbionts has been proposed for the development of bacterial bioinoculants that improve the survivor rate of the reintroduced plants. However, the isolation of the A. nitidiflorus symbiont (Ans) was not successful in the normal rhizobial growing media. PCR amplification of recA gene was performed from nodule DNA and showed that the putative Ans inhabiting the A. nitidiflorus nodules probably correspond to a Mesorhizobium sp. Two different strategies are carried out to overcome the non-culturable status of Ans: (1) direct isolation from Cartagena soils and (2) indirect isolation using relative legumes as alternative hosts. | |
