An integrated optical biosensor is used for label-free detection and a competitive limit of detection is obtained. The purpose sensor is fabricated by contact lithography technique [1] and a bio-compatible multilayer interferometric transducer is created. The resulting biosurface makes the immobilization of any possible bioreceptor easier and allows a vertical optical characterization of the biosensor. In this case a label-free detection of protein S100A6 related with dry eye disease is performed. To accomplish the S100A6 detection, in first place only 3 µL of protein A is incubated. Through this linker, a desired orientation of the antibody (bioreceptor) can lead to a better recognition. After protein A , 3 µL of 20 µg/mL antiS100A6 solution is incubated. The biofunctionalization process finishes with the incubation of 3% BSA solution as blocking agent to avoid unspecific recognition. Finally, the detection of the S100A6 is performed using different concentrations solutions of 3 µL. As a result, the recognition curve shows that the proposed biosensor is capable to detect the protein in a detection range from ng/mL to µg/mL with very low sample volume. | |
