Diversity of type six secretion system (T6SS) in rhizobia

The bacterial Type VI secretion system (T6SS) is a molecular nanosyringe able to inject effector proteins directly into prokaryotic or eukaryotic cells. Most of the proteins related to T6SS (effectors, components of the nanosyringe and regulatory proteins and chaperones) are frequently encode by clustered genes (Lien and Lai, 2017). Recently it has been shown that this system has a positive effect on the Rhizobium-beans symbiosis (Salinero-Lanzarote et al, 2019). Many of the proteins of the nanosyringes are conserved between different bacteria and orthologs have been identified from the proteomes deduced from sequenced rhizobia. In this work, a phylogenetic analysis of the rhizobial T6SS was carried out using the amino acid sequence of TssB, a core element of the contractile tube of the nanosyringes. The results showed 4 main groups. Group 1 includes species belonging to the Rhizobium genus such as R. leguminosarum and R. etli. Group 2 contains two subgroups, the first consisting of amino acid sequences of genera Rhizobium, Neorhizobium, Sinorhizobium, Ochrobactrum and Azorhizobium. The second subgroup included the TssB from Methylobacterium nodulans, R. etli CIAT552, R. phaseoli Ch24-10 and Paraburkholderia phymatum STM815. It can be emphasized that the last three strains have two T6SS, and their second TssB are located in group 4. Group 4 is the most diverse and covers also strains from Mesorhizobium, Bradyrhizobium, Devosia, Bosea and Cupriavidus genera. Group 3 is represented only by strains of the genus Bradyrhizobium. On the other hand, in this work we compare the genetic organizations of the different groups with emphasis on the conserved genes and especially on the possible effectors where the diversity is very large both among TssB groups and among rhizobial strains.
