Micropropagation isawidelyusedtechniqueinhops(Humulus lupulus L.). However, to thebestofourknowledge,thegeneticandepigeneticstabilityofthemicroplants has neverbeentestedbefore.Inthepresentstudy,twohopaccessionswere established in vitro and micropropagatedfor2years.Thegeneticandepigenetic stability ofthe in vitro plants wasanalyzedwithseveralmoleculartechniques: random amplifiedDNApolymorphism(RAPD),retrotransposonmicrosatellite amplified polymorphism(REMAP),andmethylation-sensitiveamplificationpoly- morphism (MSAP).Nogeneticvariationamongcontrolandtreatedplantswasfound, even after12cyclesofmicropropagation.Epigeneticvariationwasdetected,first, when fieldand in vitro samples werecompared.Nearlya30%ofthedetected fragments presentedthesamepatternofalterationsinallthevitroplants.Second, lower levelsofepigeneticvariationweredetectedamongplantsfromthedifferent subcultures. Partofthisdetectedvariationseemedtobeaccumulatedalongthe12 sequential subculturestested. | |
