Discovering Transcriptional Control of Drought Tolerance in Common Vetch (Vicia sativa L.)?

The loss in crop yield due to climate change and associated drought is one of the main stresses that limits crop production, including the legume common vetch (Vicia sativa L.). Vetch is a forage grain legume of high protein content. Its high nitrogen fixation ability makes its cultivation suitable in sustainable agriculture systems. Genetic resources collections are essential to provide genetic variability for plant breeding programs. A screening of a common vetch core collection, in which we have analyzed agromorphological, physiological and biochemical parameters directly associated to drought response, has allowed us to identify potential drought tolerant and drought sensitive candidates. To better understand the genetic mechanism involved in drought response in common vetch we analysed transcriptomic differences in two different varieties. A total of 1332 transcripts were commonly deregulated in both varieties in drought condition. To obtain a better knowledge of the adaptive response to drought stress in common vetch, we completed our transcriptomic studies analysing the genes, and therefore the pathways, involved in the differential regulation between drought tolerant or sensitive varieties, by comparing DEG (Different Expressed Genes) between these varieties. A total of 2646 transcripts are DEG between sensitive and tolerant candidate varieties both, in watered and drought conditions. Our transcriptomic analyses revealed important differences in the expression of genes involved in redox homeostasis, cell wall modifications and stress-response. The presence of polymorphic variants in these DEGs has also been analysed. This information will be essential for the design of molecular markers potentially associated with drought. A total 59 high-quality SNPs and 54 SSR were found in 36 and 52 DEGs, respectively. These transcriptomic analyses were completed with different physiological studies (osmolytes levels, stomatal opening, epicuticular waxes, root architecture?). Taken together, our studies will provide valuable resources that contribute to (1) understanding the molecular mechanisms of drought response in common vetch, (2) analysing the adaptive mechanisms of drought tolerance and genomic differences between tolerant and sensitive varieties, (3) identification of drought candidate genes (4) developing functional molecular markers (SSR and SNP) associated with an agronomically important trait (drought tolerance) and further advancements in marker- assisted selection (MAS
