We have studied the stability along the encapsulation-dehydration cryopreservation process (pre-culture, encapsulation, dehydration, freeze-treatment and recovery), in order to discriminate variation associated with each step. In this new study, besides RAPDs markers, we have also used a highly discriminative technique, AFLP. Using both techniques, clear differences between control and cryopreserved samples have been detected. Furthermore, AFLP pattern profiles showed somaclonal variation associated to different steps along the process. The percentage of polymorphic bands obtained with AFLP markers was higher than with RAPDs, showing that AFLP technique is more powerful and effective to analyze somaclonal variation. A study of the percentage of variation found in each step of the cryopreservation process using both molecular markers techniques was carried out. | |
